Show moreThere exists a strong demand for orthotics and prosthetics (O & P) in Latin America, with approximately 2.5 million people in need of this type of care. IPRO 309 was started in the spring of 2006 with the goal of helping to make this type of care more readily available. However, there are only 50 certified, and 1500 uncertified, O & P practitioners in Latin America. In order to become a certified practitioner, a student must graduate from an ISPO accredited program. Unfortunately, there is currently only one ISPO accredited program in existence in Latin America, along with several other programs that are not ISPO accredited. In October of 2004, Centro Don Bosco (Bogotá, Colombia), Don Bosco University (San Salvador, El Salvador), and the Laboratorio Gilete (Bogotá, Colombia) signed an agreement to establish the first accredited O & P education program in Colombia. Since then, Centro Don Bosco has allotted 3,500 square feet of space for the thriving faculty, classrooms, manufacturing training, and vocational workspace that are necessary for an ISPO accredited program. There are three levels of accreditation according to ISPO standards. Category III involves the design and manufacturing of orthotic and prosthetic devices, Category II includes the fabrication of the devices as well as direct patient care, and Category I includes production, treatment for patients, and research and development. The equivalent to ISPO in the United States is the American Board of Certification in Orthotics and Prosthetics (ABC). Because it is independent from the ISPO, the ABC standards of accreditation will need to be taken into account in order to carefully cross cultural and national boundaries.

Show moreIn myocardial perfusion imaging using single photon emission computed tomography (SPECT), gated acquisition is often used in order to deal with blur caused by cardiac motion in the resulting images. While this can provide useful information about the myocardial function, it also inevitably leads to reduced signal-to-noise ratio in the acquired data due to gating. In this work, we aim to investigate and evaluate image reconstruction methods for improving the quality of the reconstructed images in cardiac gated SPECT imaging. First, we propose a spatio-temporal (aka 4D) reconstruction procedure for gated images based on use of discrete Fourier transform (DFT) basis functions, wherein the image activity at each spatial location is regulated by a Fourier representation along the gate dimension. The gated images are then reconstructed through determination of the coefficients of the Fourier representation. We explore two different reconstruction algorithms, one is a penalized least-square approach and the other is a maximum a posteriori approach. Our simulation results demonstrate that use of DFT-basis functions in gated imaging can improve the accuracy of the reconstruction. While in gated imaging the tracer distribution is traditionally treated as constant, a recent development is gated dynamic imaging where the goal is to obtain an image sequence from a single acquisition which shows simultaneously both cardiac motion and tracer distribution change over the course of imaging. In this work, we further develop and demonstrate a fully 5D (3D space plus time plus gate) reconstruction procedure for cardiac gated, dynamic SPECT imaging, where the challenge is even greater without the use of multiple fast camera rotations. We develop and compare two iterative reconstruction algorithms: one is based on the modified block sequential regularized EM (BSREM-II) algorithm, and the other is based on the Bsplines algorithm. Our simulation results demonstrate that the 5D reconstruction xiii procedure can yield gated dynamic images which show quantitative information for both perfusion defect detection and cardiac motion. Based upon the success of 5D reconstruction, we also study the saliency of 5D images for detection of perfusion defects. We explore efficient ways for characterization and visualization of information pertinent to perfusion defects in a 5D image sequence. We apply various metrics to quantify the degree to which perfusion deficits can be detected. We show that these metrics can be used to produce new types of visualizations, showing wall motion and perfusion information, that may potentially be useful for clinical evaluation. Finally, with the ultimate goal of effective detection of lesion defect for clinical use, we also investigate a direct reconstruction approach to determine a sequence of gated, kinetic parameter images from a single acquisition, which can provide information simultaneously for both tracer kinetics and wall motion. To combat the greatly under-determined nature of the problem, we apply smoothness constraints to exploit the similarity both among the different gates and among the local spatial neighborhood. The parameter images of the different gates are then determined jointly using maximum a posteriori estimation from all the available image data.

Show moreAtorvastatin (ATO), a widely used statin for lowering cholesterol was examined for its chemopreventive/therapeutic activities in prostate cancer cells. We found that ATO inhibited cell proliferation and induced autophagy in PC3 prostate cancer cells, as marked by significant induction of an autophagy marker LC3-II. Using Taqman RT-PCR technique, we also found that ATO treatment for 24h and 48h consistently up-regulated miR-182 in PC3 cells. However, adding geranylgeraniol (GGOH) to the culture media reversed the effect of ATO in regulating miR-182, suggesting that ATO up-regulates miR-182 through inhibition of geranylgeranyl biosynthesis. Overexpression of miR-182 in PC3 cells significantly decreased cell proliferation by about 36% (MTT assay), while knock-down of miR-182 stimulated cell proliferation by about 43% (MTT assay). In screening for miR-182 target genes, we found that Bcl2 and p21 are potential miR-182 target genes; Bcl2 was significantly down-regulated by ATO at both mRNA and protein levels and miR-182 knock-down up-regulated Bcl2 protein; p21 protein expression was positively correlated with alteration of miR-182 expression levels in PC3 cells. Through screening database of miR-182 target genes from TargetScanHuman 6.2 and PicTar, we found that p21 is not the direct target gene of miR-182, so it could be regulated by miR-182 indirectly. It has recently been established that miR-182 regulation is p53-dependent, since PC3 cells are p53 negative, it is clear that ATO regulates miR-182 in a p53-independent manner. These data demonstrate that miR-182 up-regulation and Bcl2 down-regulation by ATO could be two independent events and both could be involved in ATO mediated cell proliferation and autophagy.

Show moreRenewable energy is one of the most important and widely researched topics today. It is classically defined as any form of energy coming from any naturally replenish-able source. This may include everything from solar to wind power, as well as biomass or biofuels. When considering biomass, or any (living or recently-dead) biological material, the chemical energy of the molecules is generally collected through combustion. The area of liquid fuels from biomass has especially gained much notoriety and support in recent years. This is due to the lower emissions and clean-burning nature of these fuels when compared to more traditional approaches, as well as the obvious renewable nature of the starting material. While vegetable oils or animal fats can be used as a replacement for diesel fuels, corn, switchgrass, or other grains are more widely used to produce ethanol for use in common combustion engines. Today’s E85 fuel is sold to customers with a chemical makeup of 85% ethanol and 15% gasoline. The use of solid biomass as a direct supplier of energy, however, is an area still left relatively unexplored in this growing field. In theory, and as preliminary research suggests, harvesting energy directly from solid biomass may be considerably more efficient than gathering it from its processed liquid counterpart. In fact, some studies suggest that the energy acquired from burning ethanol is up to 67% lower than is contained in the plant cellulose from which it is derived.[1] There are, however, several other factors besides energy projections to consider when looking at the economic and market viability of such an approach. For example, one of the main advantages of liquid fuels over solid is the ease of transportation and storage. Additionally, the feasibility of developing a whole new process of biomass collection and processing must be balanced with economic and logistical constraints. This includes not only careful analysis of energy and cost balances, but also in-depth examination of all equipment, manpower and environmental limitations. IPRO 349 was established to examine these (and many more) considerations in the viability of sold fuel from biomass. Specifically, we have narrowed the scope of our research to biomass derived from corn stover (leaves and stalk left in the ground after harvesting) within the state of Illinois. Illinois was chosen because it is currently the largest producer of corn in the nation.[2] Corn stover has been shown to have an energy content of 5,290 Btu/lb. wet, and 7,560 Btu/lb. dry.[2]With such an approach, it may be possible to utilize what would otherwise be considered “waste” to produce useable, renewable energy. For the purposes of this project, cogeneration, or the simultaneous generation of both electricity and useful heat will be examined.

Show moreWe would like to develop a system that measures the applied torque at a bicycle crank set. In contrast to current solutions, we want to be able to retrofit our system to existing crank sets, obviating the need to abandon parts that the bicyclist already owns. In principle, according to preliminary tests performed at the MMAE department, this can be done using sets of quite inexpensive strain gauges. However, being able to get accurate torque measurements will require some advanced processing of signals from the strain gauges. These signals can then be transmitted wirelessly to a bicycle computer like the Edge 705 that the global positioning system corporation Garmin released this spring. There is a defined wireless protocol, called ANT+Sport, which has been developed specifically for the purpose of transmitting exercise data, such as power output or heart rate, to small computers. Chipsets and development kits for this protocol are also available commercially. Therefore, our task will be to find an optimal configuration of strain gauges that will be attached to the crank set, and to develop an algorithm to process the strain gauge data in order to isolate a signal that is proportional to the applied torque. This signal will then be transmitted to the bicycle computer for display and storage

Show moreRecreational fishing is a sizable market, generating over a billion dollars of economic output and providing well over a million American’s with jobs. There are many variations of fishing hooks available that retain the same basic shape and structure, the most popular examples being, the standard J-hook, treble hook and the new “Trokar”, which is “surgically sharpened”. However, there are many limitations associated with existing fishing hooks. One such limitation is that fishing hooks are not able to reliably hold the fish after embedding. Introducing a barb to the hook usually solves this issue, doing so however creates problems when setting the hook and furthermore, barbs can cause damage to the fish during the removal process. Another concern is that contemporary fishing hooks have problems with snagging on nearby rock formations, algae, weeds, or other vegetation in the water.
