Show moreVibrational control is an open-loop control strategy implemented by fast zero-average oscillations in the input variables of a system. It has been previously shown that vibrational control of an exothermic reaction in a CSTR, introduced via oscillations in the input flow rate, modifies the dynamic behavior of the reactor and enables its stabilized operation in the unstable steady-state region. In the present paper it is shown that simultaneous vibrations in input flow rate and concentration lead, in addition to a reduction of the amplitude of oscillations in reactor temperature and/or increase in reactant conversion. It is also shown that the phase shift between the vibrations of the two input variables affects the behavior of the system as well as the amplitude, the frequency and the form of forcing functions.

Show moreRed raspberry (Rubus idaeus L.) contains a variety of polyphenols including anthocyanins and ellagitannins. Red raspberry polyphenols absorbed in different forms (parent compounds, degradant or microbial metabolite molecules) are subject to xenobiotic metabolism in the intestine, liver, and/or kidney, forming methylate, glucuronide, and sulfate conjugated metabolites. Consistent exposure of certain polyphenols to the gut microbiota may act as prebiotic-like substances feeding the beneficial gut bacteria and changing the gut microbiome composition and function. The dichotomy between the biotransformation of polyphenols into their metabolites by gut microbiota and the modulation of gut microbiome composition by polyphenols is hypothesized to contribute to positive health outcomes. The present study examined the regular consumption of red raspberry purée (RRB) and/or fructo-oligosaccharide (FOS) on gut microbiome composition and subsequent bioavailability of red raspberry polyphenols in healthy volunteers. An 8-week pilot study, including two 4-week chronic treatments and 3 postprandial days, served as a feasibility study and mechanism to collect multiple biological specimens for method development. An ultra high-performance liquid chromatography (HPLC) coupled with electrospray ionization quadrupole time of flight (QTOF) and triple quadrupole (QQQ) mass spectrometer were used to identify and quantify the phenolic compounds in red raspberry purée, plasma and urine samples. Fecal samples were used for the metagenomic study. The sequencing of the 16S ribosomal RNA gene was utilized to study the gut microbiome composition. The red raspberry purée contained 148.55 ± 5.43 mg/100 g fresh weight (FW) polyphenols. Chronic RRB and/or FOS exposure influenced gut microbiome composition: at the phylum level, 4-week FOS (8 g/d), RRB (125 g/d), or FOS plus RRB (8 and 125 g/d, respectively) exposures all decreased Firmicutes and increased Bacteroidetes; at the genus level, 4-week FOS, RRB, or FOS plus RRB exposures all boosted Bacteroides and diminished Blautia; and the increased Akkermansia was only observed after RRB exposure. Chronic RRB and/or FOS exposure also altered the observed RRB polyphenol metabolites: the parent anthocyanins, such as cyanidin 3-O-sophoroside, were lower in plasma and urine after adaptation to RRB, while the production of urolithin A glucuronide (the main microbial-derived metabolite of ellagitannins) increased after FOS, RRB and FOS plus RRB exposure; an effect hypothesized to be related to the altered composition and metabolic activity of the gut microbiota. Overall, these data suggest chronic RRB and/or FOS exposure influenced gut microbiome composition and subsequently increased gut microbial metabolites.

Show moreA bifunctional ligand that can rapidly form a stable complex with a metal while conjugated to a tumor-targeting moiety is essential for targeted therapy and imaging of cancer. New bifunctional chelators in the NETA and DEPA series were developed for targeted radiation cancer therapy. The chelators showed rapid complexation kinetics with 90Y and 177Lu, and the corresponding radiolabeled complexes were stable in vitro and in vivo. Trastuzumab, Panitumumab, and c(RGDyK) conjugates (targeting HER-2, HER-1, and integrin αvβ3, respectively) of the novel bifunctional ligands were prepared. The conjugates displayed fast complexation kinetics with 90Y, 177Lu, and 205/6Bi, and the antibody and peptide conjugates labeled with 90Y, 177Lu, or 205/6Bi exhibited excellent stability and tumor targeting in mice bearing human colorectal cancer (LS-174T). Transferrin (Tf) and bile acid (BA) conjugates of novel chelators in the NE3TA series were prepared for targeted iron chelation therapy (ICT) and positron emission tomography (PET) imaging applications of cancer. Tf and BA conjugates of NE3TA had high cytotoxicity on HeLa, HT29, and PC3 cancer cells. The Tf-N-NE3TA and BA-N-NE3TA also displayed rapid complexation kinetics with 64Cu, and the Tf and BA conjugates of N-NE3TA labeled with 64Cu were stable and had high cellular uptake in HeLa and PC3 cancer cells. Tf-N-NE3TA was further conjugated with a fluorescence dye (Cy5.5) to develop targeted theranostic agent of cancer. Tf-N-NE3TA-Cy5.5 showed great radiolabeling efficiency with 64Cu, and the Tf-N-NE3TA-Cy5.5 labeled with 64Cu exhibited high cellular uptake in HeLa, HT29, and PC3 cancer cells. In summary, we have developed new bifunctional ligands having excellent chelation chemistry of 90Y, 177Lu, 205/6Bi, and 64Cu, and the tumor-specific antibody and peptide conjugates of the novel bifunctional chelators have a great potential for targeted cancer therapy and imaging as well as theranostic applications of cancer.

To build a powerful machine while at the same time satisfying reliability requirement and energy constraint, HPC scientists continue to seek a better understanding of system and component behaviors. Toward this end, modern systems are deployed with various monitoring and logging tools to track reliability and energy data during system operations. Since these data contain important information about system reliability and energy, they are valuable resources for understanding system behaviors. However, as system scale and complexity continue to grow, the process of collecting system data to extracting meaningful knowledge out of overwhelming reliability and energy data faces a number of key challenges. To address these challenges, my work consists of three parts, including data preprocessing, data analysis and advanced modeling.

Show moreHow can urban infrastructures be reinterpreted based on their social potential? The thesis discusses the value that infrastructures can have in relation to mobility and quality of life, in the context of the urban project, and proposes an example of how underutilized spaces, that pertains to an outdated model of the city, can be re-configured as adaptable and dynamic infrastructures. The Boulevard System of Chicago emerged during the process of “beautification” that the “City Beautiful” movement fostered in the 19th century. From being the “Emerald Necklace”, as it was known, the Boulevard System became today a fragmented infrastructure that mirrors the socio-economic segregation of Chicago neighborhoods. Within the archipelago of different urban realities of the city, the Boulevard System represents today an under-utilized urban space that reinforces the already existing urban thresholds between different urban communities. The goal of the research is to re-think the Boulevard System as a new transportation corridor, able to interact with other urban systems. The proposal for the boulevards is part of an Infrastructure Plan for Chicago that aims to propose ways inwhich different actions on infrastructure can revitalize the City. The idea is to re-activate the boulevard through a series of interventions, articulated around the re-connection of the fragmented system, as a new “ring” infrastructure, and the engagement with the surrounding specificities. The proposed Bus Rapid Transit system should offer an experience through the parks in Chicago and support its reactivation through the promotion of activities and events. Furthermore, the creation of a hub around the Garfield Green Line station is meant to propose a more engaging transit experience around the city of Chicago.

Show moreDepth estimation plays an important role in three-dimensional computer vision area. Its recent development focuses on real-time application. To be able to provide depth map for real-time applications like pedestrian detection and intelligent vehicles, three challenges must be overcome: 1. Real-time processing speed; 2. Insensitive to brightness change; 3. Clear boundaries and smooth surface. The thesis first describes the major steps of depth estimation, then many commonly used methods are reviewed. From them and other related real-time method, we found that the iteration based semi-global matching with mutual information has much potential to be improved. Based on that, the thesis proposed a method with two layers to provide depth map for pedestrian detection. The low resolution layer’s task is to produce a coarse depth map as quick as possible, then it helps to produce accurate mutual information distribution without iterations. The Full-resolution layer applies semi-global matching with two optional simplification schemes to speed up. The proposed method is implemented on both CPU and GPU. Experimental results and evaluation shows that it has great insensitivity to brightness changes and achieves real-time processing speed while maintaining a comparable performance with state-of-the-art real-time depth estimation methods.

DFIT uses the Item Parameter Replication (IPR) algorithm to generate statistical cuto s, which relies on several simpli cations have not been formally tested. This study examines these assumptions by comparing the IPR algorithm with an extended, unsimpli ed procedure intended to represent results observed in practice. Monte Carlo simulation was used to compare cuto s between the two methods under a number of conditions. Di erences between the IPR and extended cuto s indicate that Type I error rates are conservative at the test level and in ated at the item level. Mean group di erences resulted in massive in ation. Sample size and test length had only minor inuences on error rates.

Show moreThe Dyadic Adjustment Scale (DAS) is the most widely used measure of dyadic adjustment for individuals in committed relationships (Spanier, 1976). However, little research has focused on whether DAS measures the construct of dyadic adjustment in a way that is equivalent and unbiased across genders. The current study utilized matched moderated regression (MMR) to assess each item of the DAS to detect if gender differences in the relationships between item responses and the construct being measured are due to (a) factors other than the construct and (b) differences in the construct. Archival data were acquired from a previously published study (Eddy, Heyman, & Weiss, 1991). The sample was very large (N =3322) so it was divided into two replication groups in such a way that no couples were included in the same group. A number of statistically significant differences were found on items in both replication samples; however: (1) many of these items were not consistent across replicate groups; (2) Even when there was a consistent gender difference in both replication groups, the magnitude was small; (3) When all of the differences were summed across items, bias in total scale score was minimal because the direction of the biases differed across items and so cancelled out and; (4) A small gender difference may exist in preferences for demonstrations of affection. Findings suggest that there are not substantial gender bias or scale equivalence problems with the DAS. The construct of dyadic adjustment was similar in men and women. These findings are congruent results from the recent study of South and Kruger (2009) on gender differences in the factor structure on the DAS and lend support to the valid use of the DAS in studies of dyadic adjustment.

Show moreThe purpose of this study was to investigate the potential effects of technology and personality on training using a computer based learning environment designed to improve learning through technology-delivered instruction. Data were collected from a sample of undergraduate students (N = 98), recruited from three universities (two in the US and one in Canada), who participated in both experimental days. Condition had no effect on learning outcomes, while Conscientiousness was found to be significantly correlated with academic performance. In further analyzing the relationship between personality and learning outcomes across time using a latent growth curve model results showed non-significant conclusions for the direction of the relationship for each of the Big Five. Discussion of the results and implications for practice and future research are provided. In conclusion, this study provides a basis for considering personality characteristics when designing computer-based training interventions in organizational settings.

Show moreThe production of weak vector bosons in association with jets and their properties are intensively studied using proton-proton collision data collected with the CMS detector. In this proceedings, we will show the cross section measurement of the electroweak production of a Z boson in association with 2 jets at √s = 8 TeV, including a new data-driven approach and studies on the hadronic activity in the region between the two tagging jets. The azimuthal correlations and event shapes in Z + jets events at √s= 7 TeV are also presented.
