Show moreCancer continues to be the second leading cause of death in the world with one in two individuals suffering from some form of cancer within their lifetimes [1]. The main objective in most cancer therapies is to achieve a complete resection of the cancerous cells, which in surgical approaches is evident from the diagnosis of positive resection margins during postoperative pathology. However, pathological analysis is performed after the patient has been discharged. Thus technologies capable of identifying tumor margins in resected samples intraoperatively could substantially reduce the psychological and economical burden of “call-back” surgeries. As a step in this direction, a wide-field Raman spectroscopy system was employed, which has the capacity to discriminate between tumor and healthy tissue through a “dual-tracer” staining approach within the surgical window. The approach entails dipping the resected tissue in two tracers: one targeted to a cancer specific cell-surface receptor, and another left “untargeted”. In this experiment human glioma (U251) and human epidermoid (A431) tumors were grown subcutaneously in ten nude athymic mice. The data obtained from the Raman spectra of different tissue specimens were then analyzed through tracer kinetic modeling to quantify cancer molecule abundance as a surrogate of cancer presence. The salient finding in this study was that the model was able to predict the expected differences in EGFR concentration amongst the healthy tissue, U251, and A431. Kinetic model estimation of the ratio of EGFR concentration in U251 and A431 cell lines were in the ratio of 2:1, which correlates, with the expected ratio of EGFR concentration determined by flow cytometry which was close to 4:1. This technology not only promises to xii highlight tumor margins on a molecular level but can also be leveraged to analyze the molecular environment of the tumor to guide adjuvant precision therapies. Tracer kinetic modeling approaches were also applied to a guided precision therapy called Photodynamic therapy. Not only could the analysis of data provide parameter values for the tracer kinetics but an approach was also developed which with only a few initial data points could predict the time optimal for therapy in advance of the drug concentration maximum.

Show moreThis study evaluated the efficacy of three pulsed light systems (1.8, 3 and 100 Hz) for inactivation of Salmonella Montevideo on the surface of black pepper. The impact of frequency, treatment time and distance from the lamp on the rate of inactivation was studied. A reduction of 4.21 1og10 CFU/g was achieved with a 35 s treatment at 16.6-cm from lamp at 1.8 Hz. Similarly, reductions of 4.72 CFU/g and 4.70 CFU/g were obtained at 14.1-cm after 35 s treatment at 3 Hz and 100 Hz, respectively. Temperature measurement of packed and unpacked black pepper indicated that packaging material absorbed a small amount of energy. Measurement of surface and internal temperature suggested that temperature below the surface increased far less than surface temperature. For instance, temperature on the surface of the black pepper can go up to 200 °C as measured by the infrared camera within 30 to 40 s treatment. However, the temperature at 1 mm below the surface was up to 100 °C depending upon the distance from the lamp. The color of the black pepper did not change after pulsed light treatment (p>0.05). These results suggest that pulsed light can potentially be used to inactivate pathogens on black pepper surface without adversely affecting its quality.

Show moreConstruction managers use scheduling methods such as the critical path method (CPM) and linear scheduling methods (LSM) to improve the outcome of their project. CPM is a popular scheduling method that is mostly based on precedence relationships and the identification of the longest path through a network. LSMs are based on a continuous flow of resources and are used mostly on projects that exhibit repetitive characteristics. Despite the many obvious advantages of CPM, its use in construction has been limited. LSMs are much less frequently used than CPM, although the general consensus is that LSMs are quite effective in projects composed of activities of repetitive nature. Understanding the reasons why these methods are not used as extensively as expected could improve their level of acceptance in the construction industry. The link between construction scheduling methods and the tasks expected to be performed by schedulers has been an on-going concern in the construction industry. The link between construction scheduling methods and the capabilities of the scheduling staff has also been an important issue. This study proposes a combined task/staffing-technology fit model to understand why CPM and LSM are not used as extensively as expected in construction scheduling: (1) a task-technology fit model that aims to measure the extent to which a construction scheduling method functionally matches the tasks expected to be performed by the scheduling staff; (2) a staffing-technology fit model that aims to measure the extent to which a construction scheduling method matches the staff`s experience, know-how and capabilities. The models that are proposed are an answer to the lack of proper instruments for evaluating the extent to which scheduling methods are used in the industry. A questionnaire survey was administered to professionals listed in the directory of the Construction Management Association of America (CMAA) who have many years of experience in construction scheduling. Statistical analyses were performed to determine statistical significance of the differences between the level of agreement concerning task-technology fit and staffing-technology fit for CPM and LSM applications. Findings indicate that there are significant differences between the opinions of CPM users and LSM users. This means that task-technology fit is much different in CPM and LSM applications. Similarly, staffing-technology fit is also quite different in CPM and LSM applications.

Show moreThe population of patients with end stage renal disease (ESRD) is growing at a rate higher than the global population. The only viable treatment for these patients is a kidney transplant. However, in the absence of a suitable kidney donor, renal patients are left with hemodialysis as a renal replacement therapy. Hemodialysis is facilitated through arteriovenous fistula (AVF), and the particular interest in this investigation is brachiocephalic fistula (BCF). The survival of dialysis patients depends on maintaining patency of fistula over a prolonged period of time. The extreme hemodynamic environment that is created by BCF triggers the onset of neointimal hyperplasia (NH) in most renal dialysis patients, which leads to access failure via stenosis. This is because the hemodynamics in AVF are well outside the normal physiological range. Computational fluid dynamics (CFD) along with shape optimization allows for the study of the hemodynamic parameters such as wall shear stress that have been shown to be detrimental in the future occurrence of cephalic arch stenosis. In this study, CFD modeling and identification of hemodynamic patterns was possible in three dimensions due to advanced post processing of IVUS patient-specific geometries. A method utilizing 3D CFD and shape optimization has been developed to implement Miller’s banding method used in clinical practice to evaluate its impact on WSS and onset of neointimal hyperplasia. The level of banding represented by a constriction is in fact a patient specific value and is not a trivial solution of minimum flow rate necessary to conduct hemodialysis; hence suggesting that even restoring the inlet velocity to the velocity pre-fistula creation might not reduce incidence of cephalic arch stenosis. The findings of this study support the previous hypothesis that non-homeostatic WSS distributions trigger neointimal hyperplasia and resulting venous stenosis. The important outcome is that the presented computational framework allows for evaluation of Miller's banding method for reducing the blood flow rate via surgical constriction and identification of a patient-specific banding level that restores the WSS to the normal physiological range.

Show moreStereo Vision is one of the fundamental problems of computer vision and one of the oldest and heavily investigated areas of 3D Vision. Recent advances in stereo matching methodologies have allowed researchers to develop several stereo vision techniques. But it has been observed that most of these methodologies mainly focus on only one aspect, either the execution time or the quality of the generated depth map. While attempts to maximize the quality of the depth map have been successfully achieved, it is however observed that the execution time has increased signi cantly, thereby rendering them un t for real time implementation. In this thesis, an attempt to solve the above said problem is presented, in which we combine the e ects of two important stereo matching concepts, one attempting to increase the depth map quality, while the other being that of having an algorithmic structure so that in future, it can be used for real-time implementation. By applying hardware-friendly methods to the framework of segment based stereo matching, the method is said to achieve a good quality depth map. This work also discusses the various challenges which it takes care of plus the study of performance compared to existing methods.

Show moreIn this research, we have explored and verified the data security and real-time performance of a Secure Wireless Health Data Management (SWHDM) system applied to personal body sensor networks. Security must be considered critical for any healthrelated information system. The SWHDM examined for this study consists of multiple wireless health-related sensors, one central data coordinator called the Wireless Intelligent Personal Communication Node (W-iPCN), a custom application designed for an Android smartphone, and a remotely located central medical server. There are two major communication channels involved in wireless data transmission. We extensively studied the body sensor data for wireless data transmission. We also focused on applying cryptographic algorithms in order to provide data confidentiality, integrity and authentication. This security assessment aims to observe the performance of data encryption and decryption, and feasibility to remotely monitor patients' health in realtime. The test result based on ECG examples is favorable towards the real-time processing requirements of the system.

Show more124 participants were recruited from thirteen undergraduate psychology courses and responded to online surveys over the course of a semester, rating their instructors' behaviors, their own behaviors and experience with their course. Results supported the hypothesized main effect of transformational leadership on students' class-engagement and satisfaction with a course. Additionally, student engagement mediated the relationship between the leadership style and satisfaction. Results also indicated that transformational leadership has an indirect effect of on two measures of student performance (final course grades and students' expected course grades) through student engagement. In conclusion, the findings of this research indicate the transformational leadership has a significant and positive role in engagement, satisfaction and learning, further supporting the value of examining instructor behavior as leader behavior.

To create suitable biological scaffolds for tissue engineering and cell therapeutics, it is essential to understand the matrix mediated specification of cell lineage. Our study examines the role of matrix properties on cellular behavior and tissue mechanics. To this end, we studied the effect of collagen type I on stem cell differentiation and its mechanical properties within a live tissue. We altered the properties of collagen type 1 by incorporating CNT. The collagen-carbon nanotube (collagen-CNT) composite material was stiffer with thicker fibers and longer D-period. We find that the enhanced mechanical and structural properties of collagen-CNT allow for rapid and efficient derivation of neural progenitors from human decidua parietalis placental stem cells (hdpPSC). Both structure and stiffness of the matrix are important determinants of neural differentiation rate. Strikingly, the collagen-CNT matrix, unlike collagen, imposes the neural fate by an alternate mechanism that is independent of beta-1 integrin and beta-catenin. The study demonstrates the sensitivity of stem cells to subtle changes in the matrix and the utilization of a novel biocomposite material for efficient and directed differentiation of stem cells. Investigation of connective tissue disorders has led to the understanding of the important role played by collagen. So far, native collagen fibers within an intact tissue have not been examined. In this study, we employed a unique approach- histochemical staining guided high-resolution elasticity mapping- to study collagen and smooth muscle in fresh vaginal wall connective tissue. The comparative study of tissues collected from healthy pre-menopausal (pre-M) and post-menopausal (post-M) women suggest that during menopause, collagen’s structure and elasticity are subtly altered. The systematic analysis enables detection of minute changes in collagen in non-fatal conditions such as pelvic organ prolapse and other genitor-urinary disorders, where the initial symptoms are subtle and multivariate and where early diagnosis will allow non-invasive interventions and reduce incidence of surgical correction for these common disorders.
