Show moreNanopore stochastic sensor using a biological protein ion channel has emerged as a powerful sensing technique due to its outstanding features such as label- and amplification-free detection, rapidness, high sensivity and selectivity. Development of a nanopore stochastic sensor for the rapid detection of anthrax can effectively prevent and control the spread of disease infection, and become extremely useful in biodefense. Two sensor design strategies were studied: one is chemistry-based detection of dipicolinic acid; and the other is genetics-based detection of anthrax lethal factor. Both approaches used in this study should find useful application in the development of nanopore sensors for the detection of other biomolecules and pathogens.

Show moreEfficiency in parking facilities has been set to low standards since self-parking was introduced in the 1950s1. As technology has developed, parking has been left in the past. Drivers are increasingly realizing the problem as they become accustomed to other technology, which increases the efficiency of their lives (eg smartphones). While driving around searching for an open lot, circling a garage for an parking spot, or wandering through garage a looking for a parked car, people have time to ponder the many improvements that should be made to the parking experience. Simply Park: IPRO 365 is taking initiative to seize the possibilities new technologies offer the parking industry. The result should be improved driver experience as well as increased efficiency and profitability of parking facilities. The system will include signs visible to passersby that display the number of vacancies within a parking facility. Once in the parking facility, additional signs will direct drivers to sections of the facility with vacancies. Colored lights above each parking spot will indicate the state of occupancy for quick reference when looking down an aisle. When a driver parks, the ticket they receive at the entrance gate will associate their ticket to the spot they parked in, so when the driver returns to the facility he/she can present his/her ticket to a kiosk which will notify him/her how to get back to his/her car. As the vehicle leaves the ticket association will be reset and displays will update to show one more spot available. Additional interfaces through mobile and internet applications will further expedite the parking process. 1National

Show moreIndividuals with spinal cord injury (SCI) are at greater risk for medical problems, and an important part of lifelong rehabilitation is the prevention of these complications. Areas of concern include pressure ulcers, lower urinary tract problems, respiratory infections, chronic pain, depression, obesity, leg spasms, bowel impaction, cardiovascular problems, poverty and social isolation. Programs that can prevent or manage these problems from the patients home are at the forefront of medicine today. Web based technology holds great promise in this area because of its interactivity, easy access, availability throughout the day, and low cost. The purpose of this IPRO is to develop a Web site or a model of a site for home-based activities and to have the site evaluated by stakeholders at Hines VA Hospital SCI Service. The seven SCI physicians and two nurse practitioners associated with this Service are very enthusiastic about meeting with students to provide their input related to any Web platforms that may be created. Specific tasks for the project include: (1) Structure of the site to incorporate such elements as: (a) One secure Home Web page with additional secure Web pages for each patient, (b) Web pages for each of a patients problems and programs for management, (c) Access by care givers to the patients Web pages for monitoring, feedback and communication with the patient; (2) A database that includes: (a) Data from structured questionnaires (already developed) related to secondary complications. Patients would be expected to periodically answer the questions associated with their problem areas; (b) Longitudinal summary reports of a patient's responses to easily show condition progress;(3) Two-way communications that includes: comment boxes for staff and patients to communicate or chat in each problem area;(4) Integrates standard home healthcare program components that include: These include stress management, exercise, and medication management. Tracking should include performance and feedback;(5) Solutions for Web access by the patients that offer possibilities: Solutions could including Web TV, a specially designed low-cost computer with Web access, or a PDA; (6) HIPPA and VA patient confidentiality.

Show moreUbiquitin-like protein 4A (Ubl4A) was identified as a housekeeping gene at X chromosome. It involves in the guided entry of tail-anchored (GET) protein pathway in which tail-anchored (TA) proteins are transported to endoplasmic reticulum (ER). However, Ubl4A also involves other functions not related to GET pathway, such as tumor suppression and DNA damage-mediated apoptosis. Up to date, the function of Ubl4A in mammals is still largely unknown. We found that either overexpression or knockdown of Ubl4A promoted cell death in cell culture system. Using an in vivo genetic knockout system, we found that Ubl4A knockout mice displayed a high neonatal mortality and had a defect in glycogen synthesis, which is mainly controlled by a key protein kinase Akt. Loss of Ubl4A resulted in the impairment of insulin-induced Akt translocation to the plasma membrane, an essential step for Akt activation. We demonstrated that Ubl4A directly interacted with actin-related protein 2/3 (Arp2/3) complex, further accelerated Arp2/3 complex-dependent actin branching, thereby bringing Akt to proximity into the plasma membrane for activation. Furthermore, we showed that Ubl4A-mediated actin branching also played important roles in other cellular activities, such as formations oflamellipodia and filopodia, macrophage phagocytosis, wound healing, and neutrophil chemotaxis. These findings provide us a new insight into understanding the roles of Ubl4A in cellular function and a molecular basis for treatment of related human diseases.

Show moreThis study presents the method in which the energy and ancillary services auction is simultaneously cleared in electricity market. By the security-constraint unit commitment model proposed in this study Independent System Operators (ISO) can determines the sufficient amount of reserve which is necessary to maintain security and reliability of the system. Before the fixed reserve requirement either equal to a percentage of the system peak load or a thermal unit with highest capacity is considered in energy and ancillary service auction in market clearing. The disadvantage of this method is high cost and insufficiency. When it is insufficient the system operator needs to committee more thermal units or does the load curtailment. At the time the fixed quantity is more than needed customers pay more although it is not necessary. Here the sufficient amount of reserve is determined based on the contingency which has been simulated. Contingencies include thermal unit outage and line outage is considered. The amount of reserves is obtained based on thermal units’ physical constraints and the rate offered in the electricity market. Then the integration of wind generation and its effects on the quantity of the reserve determination is considered. Since the wind power generation brings uncertainties to the power system we need to consider scenarios. In order to generate wind power generation scenarios we use Monte Carlo simulation. Since the number of scenarios are too much and increase the problem complexity we use Fast backward/forward scenario reduction. This problem is solved through direct optimization problem including the minimization of the operational cost as well as satisfying the network security constraints when contingency happens.

Show moreDeguelin, as a rotenoid, has emerged as an attractive pharmacophore for chemprevention due to its effectiveness for both in vitro and in vivo models. The biological activity and its structural complexity instigated the development of new synthetic technologies which could satisfy the increasing demand of deguelin, as well as broaden the diversity of the rotenoids. Here, a strategy of asymmetric acid-catalyzed annulations toward the enantioselective total synthesis of (-)-deguelin will be discussed. Several approaches were investigated to connect the chromene and phenolic moieties to create the pentacyclic rotenoid skeleton. Meanwhile, by collaborating with Professor Rajendra Mehta, several derivatives of deguelin were also made to study its bioactivity and interaction modes with specific proteins.

Show moreIn light of the global sustainability movement, several mainstream ideas and designs for low to zero carbon emission vehicles have been developed. In the case of all electric vehicles (EVs), Li-Ion batteries seem to be the favored technology due to their high energy density relative to competing energy storage technologies. Despite its superiority to other batteries, it still lacks the energy density to practically achieve comparable range to a fossil fuel powered car, and faces additional challenges in terms of refueling and infrastructure development. Therefore, technologies are needed to ease this demand and possibly supplement it. IPRO 313 has undertaken the task of using Zinc as an electrochemical fuel. The design is based on a Zinc-Air fuel cell developed by John F. Cooper of Lawrence Livermore National Laboratory (LLNL) [1]. The design allows the zinc to be provided as < 1 mm sized pellets in a saturated solution of KOH. Using Zinc as a fuel means that exhausted fuel, ZnO, can be recycled. The fact that the metal is recyclable lends this technology to requiring a minimum of new material to sustain the transportation infrastructure. Most importantly, in comparison to current refueling mechanisms, Cooper et. al showed that a battery can be refueled in less than 10 minutes [2] making it a good competitor in the current and emerging eco-friendly vehicle market of which the IPRO plans to build on. So far the IPRO has designed and constructed one prototype and conducted pressure drop experiments. The plans now are to improve the current ZAFC design and investigate the relationship between the electrolyte flow rate and the energy output. Once the single ZAFC has been tested and is providing the output needed, several more will be constructed and put together in an array to supplement the primary battery pack used to provide instantaneous power to the electric engine. Subsequently, the team will begin working on the staple of the project which is to design a mechanism which will refuel the ZAFC array by replacing the old electrolyte and zinc solution with a fresh electrolyte and zinc to all cells.

Show moreMany High-Performance Computing (HPC) applications spend a significant portion of their execution time in accessing data from les and they are becoming increasingly data-intensive. For them, I/O performance is a significant bottleneck leading to wastage of CPU cycles and the corresponding wasted energy consumption. Various optimization techniques exist to improve data access performance. However, the existing general-purpose optimization techniques are not able to satisfy diverse applications' demands. On the other hand, the application-specific optimization pro- cess is usually a difficult task due to the complexity involved in understanding the parallel I/O system and the applications' I/O behaviors. To address these challenges, this thesis proposes an application-aware data access optimization framework and claims that it is feasible and useful to utilize applications' characteristics to improve the performance and efficiency of the parallel I/O system. Under this framework, an optimization may consist of several basic but challenging steps, including capturing the application's characteristics, identifying the causality of I/O performance degra- dation, and delivering optimization solutions. To make these steps easier, we design and implement the IOSIG toolkit as an essential system support for the default par- allel I/O system. The toolkit is able to pro le the applications' I/O behaviors and then generate comprehensive characteristics through trace analysis. With the help of IOSIG, we design several optimization techniques on data layout optimization, data reorganization, and I/O scheduling. The proposed framework has significant poten- tial to boost application-aware I/O optimization. The results prove that the proposed optimization techniques can significantly improve the data access performance.

Show moreThe main problem the IPRO addressed was that of insider misuse. Contrary to popular belief, the largest threat facing companies on a technology level is not intrusion, but rather insider misuse. Insider misuse is a relatively new area of research in the field of Computer Science, which is one of the reasons it’s a relatively unheard of problem. In addition to that, due to intense media bias towards intrusion, people are blissfully unaware of the problem of insider misuse. Ethically, it’s an interesting problem. Companies already closely monitor user activity, but any infringement on perceived privacy in the workplace generally sends everyone flustering left and right. The IPRO had to personally evaluate if we felt the necessary invasion of privacy was worth the overall result which is a more secure and profitable business. Companies can decide which approach they would like to take, but it’s hard to believe that a company would choose one that wouldn’t help to prevent a loss of money.

Show moreIPRO 307 is following the path of its predecessors in order to help improve the shipping transportations and facilities in Kankakee County, IL. While there have been other IPRO teams that have contributed to the modernization of the intermodal system at MI-Jack Shipping Company, our sponsor is now looking for us to further their research in this field. They would also like us to further the research in high speed intermodal along with a viaduct enhancement system. One of our tasks is to select a location in Kankakee County to place the intermodal system. We have decided to break this into two locations with one being in-city and the other being out-of-city. Kankakee County has been selected because two major rail lines pass through it. There is also a highway that runs through the center of it. Therefore, it is an ideal 2 location for an intermodal yard. Also, there is a wind farm being built in Kankakee, so we will be able to save energy by thinking green. In our selection process for a location, we may need to relocate some people. Therefore, our plan is to have affordable housing at our out-of-city location. We will design this affordable housing along with the warehouses and storage facilities, which go along with the design of an intermodal yard.

Show moreIPRO321, Enhancing Reliability and Performance of a Paper Shredder, is focused on designing a better, more reliable paper shredder without increasing cost. The objectives of this group, as set forth by the sponsor, Seth Lewis of the Manhattan Group, are: 1. To determine the amount of force needed to cut 6, 8, and 10 sheets of 20 lb. paper taking into account the length and width of the shred 2. To design an efficient gear train that optimizes the number of gears and minimizes the motor size 3. To develop acoustical sound dampeners to reduce the amount of noise created during the shredding process by 10 decibels. The team will seek to accomplish these goals through the use of computer simulation, extended research, and laboratory testing and error. By the end of the semester the team anticipates having developed a mathematical model that will allow for the minimization of the amount of force used to shred various amounts of paper, to have built a gear train that works better and lasts longer, and to have noticeably reduced the amount of noise created by the shredder. The IPRO Team’s goal is to implement all of these changes to create a shredder with improved performance, reliability, and life span without increasing cost.
