Show moreIIT has taken part in the smart grid initiative by converting the existing radial distribution network into a looped network to increase the reliability of the system as part of the ’Perfect Power’ project. A total of 7 loops have been planned with the help of fast fault clearing relays (’Vista Switches’). As part of the project, the cable is modeled using the various impedance calculators and was also done using the PSCADrsoftware. With the obtained impedance values from the impedance calculators, the cables were analyzed under various outage/contingency cases. They include base case power flows of the old IIT distribution network as well as the Loop 3 of the perfect power project implemented and open circuit analysis and short circuit analysis of the Loop 3 alone. Also, with the help of the fault current values from the short circuit study we can analysis the relay settings and operations and review the later for the newly installed relays. And finally, the cable fault classifier was designed using the Artificial Neural Network (ANN) method in MATLABrand PSCADrsoftware. Here we model the cable using the impedance calculated and collect the voltage and current samples from the two ends of the cable which is given as input to the ANN classifier and the same is tested for accuracy.

Show moreThis thesis focus on examining the relationship between bid price and claims in public construction projects in order to propose the alternative bidding strategy and help owner minimize actual completion cost and gain the best value for their construction projects. The investigation on the literature of bidding methods, claims and their relationship guided us to come up with our hypotheses on building up an equation that quantitatively expressing the reverse correlation of bid price and claims. In order to archive our goal, data from 30 public construction projects were collected, and the statistic method and data analysis are utilized. Finally the equation to estimate the amount of claim from the bid price, the engineer’s estimate, and the spread between the lowest two bids are established through regression model. Even though some limitations are remained, the results from this study strongly recommend owners to prefer a contractor that offers the lowest bid and potential claim, rather than select the contractor using the traditional process of choosing the lowest bid.

Show moreVibrational control is an open-loop control strategy implemented by fast zero-average oscillations in the input variables of a system. It has been previously shown that vibrational control of an exothermic reaction in a CSTR, introduced via oscillations in the input flow rate, modifies the dynamic behavior of the reactor and enables its stabilized operation in the unstable steady-state region. In the present paper it is shown that simultaneous vibrations in input flow rate and concentration lead, in addition to a reduction of the amplitude of oscillations in reactor temperature and/or increase in reactant conversion. It is also shown that the phase shift between the vibrations of the two input variables affects the behavior of the system as well as the amplitude, the frequency and the form of forcing functions.

Show moreA method and system for sensing or characterizing a biomarker, such as a proteolytic enzyme or nucleic acid. The system comprises a nanopore sensor to determine a current modulation of a sample including a biomarker, and a predetermined substrate or nucleic acid probe current modulation signature for comparison to a current signature from the nanopore sensor. The nanopore sensor includes a nanopore membrane between two fluid compartments, and a power supply in electrical contact with the membrane to provide an electric potential difference between the fluid compartments. A detector is used to detect an electrical current through the nanopore as the polypeptide substrate, or components thereof, transits the nanopore under an applied electric potential difference between the first and second fluid compartments. The result is a rapid, label-free method for the sensitive and accurate measurement of biomarker activity by real-time monitoring of the ionic current modulations arising from the substrate peptide-protease interactions or nucleic acid hybridization in the nanopore.

Show moreRed raspberry (Rubus idaeus L.) contains a variety of polyphenols including anthocyanins and ellagitannins. Red raspberry polyphenols absorbed in different forms (parent compounds, degradant or microbial metabolite molecules) are subject to xenobiotic metabolism in the intestine, liver, and/or kidney, forming methylate, glucuronide, and sulfate conjugated metabolites. Consistent exposure of certain polyphenols to the gut microbiota may act as prebiotic-like substances feeding the beneficial gut bacteria and changing the gut microbiome composition and function. The dichotomy between the biotransformation of polyphenols into their metabolites by gut microbiota and the modulation of gut microbiome composition by polyphenols is hypothesized to contribute to positive health outcomes. The present study examined the regular consumption of red raspberry purée (RRB) and/or fructo-oligosaccharide (FOS) on gut microbiome composition and subsequent bioavailability of red raspberry polyphenols in healthy volunteers. An 8-week pilot study, including two 4-week chronic treatments and 3 postprandial days, served as a feasibility study and mechanism to collect multiple biological specimens for method development. An ultra high-performance liquid chromatography (HPLC) coupled with electrospray ionization quadrupole time of flight (QTOF) and triple quadrupole (QQQ) mass spectrometer were used to identify and quantify the phenolic compounds in red raspberry purée, plasma and urine samples. Fecal samples were used for the metagenomic study. The sequencing of the 16S ribosomal RNA gene was utilized to study the gut microbiome composition. The red raspberry purée contained 148.55 ± 5.43 mg/100 g fresh weight (FW) polyphenols. Chronic RRB and/or FOS exposure influenced gut microbiome composition: at the phylum level, 4-week FOS (8 g/d), RRB (125 g/d), or FOS plus RRB (8 and 125 g/d, respectively) exposures all decreased Firmicutes and increased Bacteroidetes; at the genus level, 4-week FOS, RRB, or FOS plus RRB exposures all boosted Bacteroides and diminished Blautia; and the increased Akkermansia was only observed after RRB exposure. Chronic RRB and/or FOS exposure also altered the observed RRB polyphenol metabolites: the parent anthocyanins, such as cyanidin 3-O-sophoroside, were lower in plasma and urine after adaptation to RRB, while the production of urolithin A glucuronide (the main microbial-derived metabolite of ellagitannins) increased after FOS, RRB and FOS plus RRB exposure; an effect hypothesized to be related to the altered composition and metabolic activity of the gut microbiota. Overall, these data suggest chronic RRB and/or FOS exposure influenced gut microbiome composition and subsequently increased gut microbial metabolites.

Show moreA bifunctional ligand that can rapidly form a stable complex with a metal while conjugated to a tumor-targeting moiety is essential for targeted therapy and imaging of cancer. New bifunctional chelators in the NETA and DEPA series were developed for targeted radiation cancer therapy. The chelators showed rapid complexation kinetics with 90Y and 177Lu, and the corresponding radiolabeled complexes were stable in vitro and in vivo. Trastuzumab, Panitumumab, and c(RGDyK) conjugates (targeting HER-2, HER-1, and integrin αvβ3, respectively) of the novel bifunctional ligands were prepared. The conjugates displayed fast complexation kinetics with 90Y, 177Lu, and 205/6Bi, and the antibody and peptide conjugates labeled with 90Y, 177Lu, or 205/6Bi exhibited excellent stability and tumor targeting in mice bearing human colorectal cancer (LS-174T). Transferrin (Tf) and bile acid (BA) conjugates of novel chelators in the NE3TA series were prepared for targeted iron chelation therapy (ICT) and positron emission tomography (PET) imaging applications of cancer. Tf and BA conjugates of NE3TA had high cytotoxicity on HeLa, HT29, and PC3 cancer cells. The Tf-N-NE3TA and BA-N-NE3TA also displayed rapid complexation kinetics with 64Cu, and the Tf and BA conjugates of N-NE3TA labeled with 64Cu were stable and had high cellular uptake in HeLa and PC3 cancer cells. Tf-N-NE3TA was further conjugated with a fluorescence dye (Cy5.5) to develop targeted theranostic agent of cancer. Tf-N-NE3TA-Cy5.5 showed great radiolabeling efficiency with 64Cu, and the Tf-N-NE3TA-Cy5.5 labeled with 64Cu exhibited high cellular uptake in HeLa, HT29, and PC3 cancer cells. In summary, we have developed new bifunctional ligands having excellent chelation chemistry of 90Y, 177Lu, 205/6Bi, and 64Cu, and the tumor-specific antibody and peptide conjugates of the novel bifunctional chelators have a great potential for targeted cancer therapy and imaging as well as theranostic applications of cancer.
