Show moreThis thesis explores and introduces new constructions for non-adaptive group testing which are particulary important for the parameter range we encounter in real life problems. After a summary of existing results, the rst part of this thesis introduces our own constructions, the Latin Square Construction and the Column Augmented Concatenation. Both of these constructions take existing good group testing matrices to create test matrices of larger dimensions. These new matrices are easy to nd for the practical small parameter range we are most interested in. We also address and prove asymptotic results of our Latin Square Construction. In case of the Column Augmented Concatenation the asymptotic results depend greatly on the codes used for the construction. The second part of our work is to address possible ways of augmentation of the Latin Square Construction. Here we explore the di erence in augmentation based on the properties of the starting matrix. In the appendices we give tables of best matrices coming from our constructions with xed, small column weights. We also give a list of the known best 2-disjunct matrices for small row numbers.

Show morePaenibacillus strains 32O-W and 32O-Y were attempted to be transformed by electroporation with constructed plasmid pNW33N-vgb, modified to contain the vgb gene which can be expressed as Vitreoscilla hemoglobin. Only attempts with 32O-Y were successful. Transformed 32O-Y/pNW33N-vgb was grown in CDM medium with dibenzothiophene (DBT) as the sole source of sulfur at different temperatures. Dramatic variability was observed in culture at different temperatures, so only the data at 45 °C was analyzed. The growth assay showed that the 32O-Y/pNW33N-vgb strain grew slower than untransformed 32O-Y, although Gibbs assay showed improvements in utilizing ability of DBT of 32O-Y/pNW33N-vgb compared to untransformed 32O-Y. This finding indicated that genetic engineering of introducing vgb into 32O-Y may cause deterioration in cell growth rate but improvement in desulfurization activities. Plasmid pUC-vgb-M2 was transformed into E. coli DH5α. The transformed DH5α/M2-vgb was cultured along with DH5α/pUC8:16, bearing plasmid pUC8:16 that was previously constructed in our lab and can be expressed to produce wild type VHb, and untransformed DH5α. CO-difference spectra were performed with the lysed cultures for the detection of VHb expression. As a result, DH5α/M2-vgb was confirmed to lack the ability to express functional VHb.

Show moreOxidative and inflammatory stress is associated with chronic disease development and progression. Recent studies suggest that consumption of fruits and vegetables rich in antioxidant compounds attenuate oxidative stress and inflammation. Strawberries are abundant in polyphenolic compounds with antioxidant properties, which have attracted research attention for their possible health benefits. The aim of the project was to determine whether the antioxidant properties of strawberries formulated in a beverage would dose-dependently restore oxidative stress induced by a high fat/ high carbohydrate meal in insulin resistant individuals. Plasma was collected from insulin resistant individuals (n=25) every 30-60 min over 6 h after ingestion after a 650 kcal high fat/ high carbohydrate meal accompanied with a beverage containing 0, 10, 20 or 40 g freeze-dried strawberry powder. The antioxidant capacity of test beverages containing 10, 20 and 40 g strawberry powder were 9.31± 0.66, 19.10 ± 0.60 and 33.57 ± 1.75 μmol Trolox equivalents/mL based on the Oxygen Radical Absorbance Capacity (ORAC) assay. The ORAC value for the placebo (0 g) was below the detectable limits. Biologically relevant oxidative stress was determined using the ORAC assay to estimate antioxidant capacity of the plasma and oxidation of low density lipoprotein (ox-LDL) using an ELISA assay to determine the oxidative damage. Assessment of ox-LDL over 6 hr revealed a significant effect of time (p<0.0001), however no significant difference between the treatment dosages was evident (p>0.05). A marginal treatment by time interaction suggested possible time specific differences between treatments. The evaluation of antioxidant capacity of the plasma was split into assessment of the hydrophilic and lipophilic fractions. Hydrophilic ORAC results showed ix no significant differences between test beverages over 6 hr (p>0.05), including effects of time or treatment by time interactions (p>0.05). The lipophilic ORAC also did not reveal significant differences between treatments (p>0.05). Overall, it appears that beverages with different “doses” of antioxidant capacity did not result in dose-dependent changes in oxidative stress in insulin resistant individuals. There was a trend for protection of ox-LDL by the 20 g strawberry beverage revealed by differences among treatments at 240 min compared to the placebo/0 g strawberry beverage. The findings suggest that antioxidant food properties do not translate directly to plasma antioxidant capacity as measured by ORAC and alternative mechanisms are likely responsible for biological antioxidant effects of strawberry.

Show morePeptidomimetics are a broad class of structural variant synthetic mimics of antimicrobial peptides (AMPs). AMPs are natural antibiotics found in almost entirety throughout all living organisms. Although, AMPs were once thought of as promising therapeutics to combat infectious bacteria, key shortcomings such as low bioavailability, as well as high manufacturing costs have prevented them from reaching a clinical market. Peptidomimetics capitalize on the characteristics of AMPs thought to be responsible for their antibacterial activity, such as net cationic charge and amphiphilicity, while introducing key structural differences. The principal mode of antibacterial activity of AMPs is causing disruption to the bacteria cell membrane of both Gram-positive and Gram-negative bacteria types. The complexities of cell membranes with their various lipid molecule species and proteins makes it difficult to determine mechanistic details of AMPs. It is for this reason model membrane systems such as Langmuir monolayers and supported bilayers composed of the lipid species found in that of actual cell membrane are used. In the present work we have investigated the mechanism of action of different types of peptidomimetics such as N-substituted glycines (peptoids) and oligomers of acylated-lysines (OAKs) on model bacteria and cancer cell membrane systems using various experimental techniques, such as epifluorescence microscopy (EFM), atomic force microscopy (AFM), X-ray reflectivity, and grazing incidence X-ray diffraction (GIXD). From these studies structure function relationships can be obtained, which help aid in the design of new and more effective peptidomimetics. Through these investigations it was found that the substitution of hydrogen atoms for fluorine atoms in the phenyl side chains of cyclic peptoids increases antibacterial on both Gram-positive and Gram-negative model membrane systems, guanidino containing side chains compared to amino containing side chains in α-peptide-β-peptoid chimeras show greater activity on model membrane systems mimicking the cytoplasmic membrane of both Gram-positive and Gram negative bacteria. In addition, insights were gained on the anticancer mechanisms of the naturally occurring AMP magainin-2 and OAK C12K-7α8. The significance of the model membrane system being used cannot be overstated, when the overall arching goal is to bring these peptidomimetics to a clinical market. Thus, the biological relevance of the model membrane system is of great importance. It is this motivation that has lead our research group to develop a floating planar lipid bilayer that is bound to a lipid monolayer functionalized with polyethylene glycol, which to our knowledge has not been previously done. This system and the planar Langmuir monolayer that were used in this work are ideal systems for the use of high energy X-rays that are produced from synchrotron radiation facilities. From such measurements as X-ray reflectivity (XR) and grazing incidence X-ray diffraction (GIXD) molecular scale resolution of the peptidomimetic interactions with the model membrane systems can be achieved.

Show moreThis paper examines a supersonic multi-jet interaction problem that we believe is likely to be important for mixing enhancement and noise reduction in supersonic mixer-ejector nozzles. We demonstrate that it is possible to synchronize the screech instability of four rectangular jets by precisely adjusting the inter-jet spacing. Our experimental data agree with a theory that assumes that the phase-locking of adjacent jets occurs through a coupling at the jet lip. Although synchronization does not change the frequency of the screech tone, its amplitude is augmented. The synchronized multi-jets exhibit higher spreading than the unsynchronized jets, with the single jet spreading the least. We compare the near-field noise of the four jets with synchronized screech to the noise of the sum of four jets operated individually. Our noise measurements reveal that the more rapid mixing of the synchronized multi-jets causes the peak jet noise source to move upstream and to radiate noise at larger angles to the flow direction. Based on our results, we have grounds to believe that screech synchronization is advantageous for noise reduction internal to a mixer-ejector nozzle, since the noise can now be suppressed by a shorter acoustically lined ejector.

A uniform reliability is maintained for multi-girder highway bridges with various span lengths. WIM data, gathered by California, Michigan, New York, Oregon, and Illinois State Department of Transportation are analyzed. Live load effects produced by WIM data are calculated using influence lines. Expected maximum live load effects in given periods are modeled and extrapolated by Gumbel distribution. A relativecalibration formula is developed in order to derive live load correction factors for designing/evaluating continuous highway bridges at a reliability level that is in consistent with simply supported highway bridges. Computer programming is conducted for data sorting and traffic statistical analysis.

SPM can also provide information to operators on how close a process is to non-compliance to product safety limits, and carry out periodic checks of sensor accuracy at high frequency. Traditional SPM tools such as Shewhart charts are not appropriate for continuous food processes because of autocorrelation in data. Four alternative SPM techniques are presented and applied to high temperature short time (HTST) dairy pasteurization. The study attempted to achieve compliance of the HTST process operation with the recommended Pasteurized Milk Ordinance by providing a margin between the alarm limits of the monitoring chart and the safety limits. Monitoring of residuals and parameter change detection techniques ave used for monitoring processes with autocorrelated variables. Hotelling's T-2 and residuals of canonical variates techniques are used for monitoring multivariable processes.

Show moreThe heavy quark content associated with gauge boson production is of great interest to studies of QCD. These events probe the gluon and heavy-quark parton distribution functions of the proton, and also provide a measurement of the rate of final state gluon splitting to heavy favor. In addition, gauge boson plus heavy quark events are representative of backgrounds to Higgs, single top, and supersymmetric particle searches. Recent work with the CDF II detector at the Fermilab Tevatron has measured the cross-section of several gauge boson plus heavy flavor production processes, including the first Tevatron observation of specific charm process p͞p → W + c. Results are found to be in agreement with NLO predictions that include an enhanced rate of g → c͞c =b͞b splitting. Lastly, a new analysis promises to probe a lower pT (c) region than has yet been explored, by fully reconstructing D* → D^0 (Kπ)π decays in the full CDF dataset (9:7 fb^1).
