Show moreExperiments were conducted to investigate how heating only the nozzle exit boundary layer of an axisymmetric jet, with an unheated potential core, affects disturbance growth in the initial shear layer. The exit boundary layer had a minimum density ratio of 0.74, was laminar, and had a constant momentum thickness (theta) for all levels of nozzle heating used in this study. The fluctuating velocity (u') and temperature (t') in the exit boundary layer increased monotonically with increasing nozzle temperature. Low-amplitude acoustic excitation produced a more rapid growth of coherent velocity fluctuations for the heated case than for the unheated. (C) 1995 American Institute of Physics.

Show moreIn a hybrid electric vehicle (HEV) with parallel powertrain, the system can be controlled by splitting the required power between the electric propulsion machine and internal combustion engine (ICE) to meet specific goals related to fuel consumption, efficiency, performance, and/or emissions. This power splitting scenario, which is of great hybridization importance, is in fact the control strategy or energy management of the hybrid vehicle. Performance of the system depends on the control strategy, which needs to be robust, stable, reliable, and independent from uncertainties. This Ph.D. research is focused on model based control strategies, which are proposed for parallel hybrid powertrains, showing significant advantages in performance and fuel economy. If a model based control strategy is used to develop the hybrid power management algorithm, the accuracy of the model data needs to be high for proper control. Therefore, this type of management method is parameter sensitive. Implementing system identification features into this algorithm reduces the effect. As a result, the proposed controller algorithm learns the existing component parameters while operating. Furthermore, combining the base controller with an online tuner, which simultaneously optimizes the controller for current conditions, will improve the performance of the power management. In addition, this Ph.D. thesis presents a novel neural adaptive equivalent consumption minimization strategy (ECMS) and applies it to a hybrid representative sport utility vehicle (SUV) with parallel powertrain. The ECMS is a model based optimal control strategy and is based on the minimization of both fuel consumption and battery charge usage by introducing the equivalent coefficient between them. Proper operation of the controller depends on the accuracy of the model. It also depends on the correct selection of the equivalent coefficient. In this Ph.D. thesis, specific neural network structures are proposed for both coefficient selections by drive cycle recognition and for precise model building by system identification. This thesis also presents a novel fast solution method of ECMS algorithm for real time applications.

Show moreDiabetes is one of the prevalent diseases in the USA, which affects the lives of millions of people. In 2010 only, there was a total of 234,051 deaths linked to diabetes in the USA. Research related to preclinical and clinical assays are always costly and time consuming. Modeling is a helpful method to reduce the cost of clinical experiments and accelerates the discovery and improvement of new therapies. This research is focused on the development of a high performance agent-based model simulating the pathogenesis of Type 1 diabetes mellitus in pancreas. The whole immune response takes place in three compartments, pancreatic lymph node, circulation, and pancreas. A significant part of the complex interactions leading to Type 1 diabetes takes place in the pancreatic tissue. Therefore, the focus was placed on the islets of Langerhans in the pancreas, and the interaction of CD8+ T cells and Beta cells were modeled. T cell behavior was incorporated as rules in this model such as activation, migration, proliferation, apoptosis, and cytotoxicity. Likewise Beta cell death and regeneration under the T cell attack were modeled. The model is able to capture the trends of T cell and Beta cell variations during the disease progression and portrays the role of CD8+ T cells in the process. It is expected that, with the addition of other immune system and pancreatic tissue components, the model will be a valuable tool for the planning of clinical studies.

Show moreA morphologically structured model is proposed to describe penicillin production in fed-batch cultivations. The model accounts for the effects of dissolved oxygen on cell growth and penicillin production and variations in volume fractions of abiotic and biotic phases due to biomass formation. Penicillin production is considered to occur in the subapical hyphal cell compartment and to be affected by availability of glucose and oxygen. As it stands, the model provides a wide range of applicability in terms of operating conditions. The model has been tested for various conditions and has given satisfactory results. A series of glucose feeding profiles have been considered to demonstrate the capabilities of the proposed model. It is concluded that the model may be valuable for the interpretation of experimental data collected specifically for metabolic flux analysis during fed-batch cultivation because the elements of measured specific production rates are determined from measurements of the concentrations of the components and their mass balances. The proposed model may be further used for developing control strategies and model order reduction algorithms

Show moreUV irradiation can cause DNA damage, which leads to either cell cycle arrest or apoptosis. As a major transcriptional target of p53 protein in response to DNA damage, p21 protein plays critical roles in both cell cycle arrest and apoptosis. However, the specific range of UV doses and functions of p21 protein leading to the determination of cell fate in human androgen-independent prostate cancer cells has not been completely elucidated. Here, we show that low doses of UV irradiation (< 40 J/m2) induced cell cycle arrest with up-regulation of p21 protein. However, high doses of UV irradiation (> 60 J/m2) can induce apoptotic cell death as indicated by caspase-3 activation, which is also consistent with apoptotic morphological changes. Interestingly, p21 protein is degraded at early time course of high-dose UV irradiation-induced apoptosis, pretreatment of proteasome inhibitor MG132 which can block p21 degradation but partially inhibits apoptotic cell death. Consistently, similar results were obtained in both 104-R1 cells and 104-IS cells. Taken together, the results suggest that there is a narrow window of UV doses range that serves as a “switch” point, in which cells make a decision: either cell cycle arrest or cell death. p21 protein serves as a good indicator for both cell cycle arrest and apoptotic cell death post-UV irradiation in human androgen-independent prostate cancer cells. Therefore, p21 may be a potential therapeutic target in prostate cancer.

Show moreIn the recent years, anisotropic metallic nanoparticles have drawn much attention because of their unique optical properties. Particularly, gold and silver nanorods show great potential in extensive applications such as health care, food storage, drug delivery, biosensor, and cancer treatment. Among a variety of nanorods synthesis methods, the seed-mediated growth approach stands out due to its mild and convenient experimental conditions. However, there exist two defects of this method, the poor reproducibility and low yield of nanorods. In order to create nanoparticle systems of reproducible quality in high yields, a deeper understanding of the mechanism of nanorods formation as well as identifying the more important variables is necessary. In this study, the original seed-mediated growth method has been modified to promote the reproducibility and yield of nanorods. Effects of CTAB concentration and ages of growth solution have been investigated in the nanorods formation process. The results suggest that a high concentration of CTAB is not necessary for the growth process and the age of growth solution can affect the optical performance of nanorods. In a brief survey on the stability property of nanorods, it shows that the degradation rate decreases with increasing concentrations of CTAB.

Show moreThis study describes a technique that combines the benefits of focusing schlieren and phase conditioning. Focusing schlieren blurs and drops contrast of non-critical features whereas phase conditioning emphasizes periodic flow features, and their combination produces unique results. The supersonic jets that we studied produced an intense tone referred to as screech. The measured screech tone signal was used as input to the phase conditioning circuit that adjusted the strobing light source to the vertical synchronization pulse of a CCD camera. The sharp video images obtained by this technique could either be frozen or continuously swept through one period of screech to acquire a slow motion video record of the jet unsteadiness. Two cases were visualized in this study: first, an underexpanded jet from a convergent rectangular nozzle at various fully expanded Mach numbers. Second, a supersonic jet emerging from a convergent-divergent rectangular nozzle at a design Mach number of 1.4, artificially excited by impingement tones. The results of this study illustrate the usefulness of this system in visualizing oscillatory flows.

We construct a set of leading-order parton distributions with QED corrections, NNPDF2.3QED LO set, based on the same data as the previous NNPDF2.3 NLO and NNLO PDF sets. We compare this PDF set to its higher-order counterparts, we discuss its implementation as an internal set in Pythia8, and we use it to study some of the phenomenological implications of photon-initiated contributions for dilepton production at hadron colliders.

Show moreThe goal of IPRO 320 is to create an online professional network for school teachers at the kindergarten through twelfth grade level to allow for the sharing of ideas, lesson plans and other professionally relevant information. This is a continuing project which saw limited success in the previous semester in that a functioning web site was constructed based on surveys of public school teachers but it saw very little use. This semester, the planned approach is to build on the research generated last semester by performing additional market research with teachers. Following that, IPRO 320 will rework much of the previous web site by refining some of the features, adding additional ones as the research indicates and improving the website in general, both functionally and aesthetically.

Show moreThis dissertation presents automated, systematic data collection, processing, and analysis methods for studying the spatial-temporal properties of Global Navigation Satellite Systems (GNSS) scintillations produced by ionospheric irregularities at high latitudes using a closely spaced multi-receiver array deployed in the northern auroral zone. The main contributions include 1) automated scintillation monitoring, 2) estimation of drift and anisotropy of the irregularities, 3) error analysis of the drift estimates, and 4) multi-instrument study of the ionosphere. A radiowave propagating through the ionosphere, consisting of ionized plasma, may su↵er from rapid signal amplitude and/or phase fluctuations known as scintillation. Caused by non-uniform structures in the ionosphere, intense scintillation can lead to GNSS navigation and high-frequency (HF) communication failures. With specialized GNSS receivers, scintillation can be studied to better understand the structure and dynamics of the ionospheric irregularities, which can be parameterized by altitude, drift motion, anisotropy of the shape, horizontal spatial extent and their time evolution. To study the structuring and motion of ionospheric irregularities at the sub-kilometer scale sizes that produce L-band scintillations, a closely-spaced GNSS array has been established in the auroral zone at Poker Flat Research Range, Alaska to investigate high latitude scintillation and irregularities. Routinely collecting lowrate scintillation statistics, the array database also provides 100 Hz power and phase data for each channel at L1/L2C frequency. In this work, a survey of seasonal and hourly dependence of L1 scintillation events over the course of a year is discussed. To efficiently and systematically study scintillation events, an automated low-rate scintillation detection routine is established and performed for each day by screening the phase scintillation index. The spaced-receiver technique is applied to cross-correlated phase and power measurements from GNSS receivers. Results of horizontal drift velocities and anisotropy ellipses derived from the parameters are shown for several detected events. Results show the possibility of routinely quantifying ionospheric irregularities by drifts and anisotropy. Error analysis on estimated properties is performed to further evaluate the estimation quality. Uncertainties are quantified by ensemble simulation of noise on the phase signals carried through to the observations of the spaced-receiver linear system. These covariances are then propagated through to uncertainties on drifts. A case study of a single scintillating satellite observed by the array is used to demonstrate the uncertainty estimation process. The distributed array is used in coordination with other measuring techniques such as incoherent scatter radar and optical all-sky imagers. These scintillations are correlated with auroral activity, based on all-sky camera images. Measurements and uncertainty estimates made over a 30-minute period are made and compared to a collocated incoherent scatter radar, and show good agreement in horizontal drift speed and direction during periods of scintillation for cases when the characteristic velocity is less than the drift velocity. The methods demonstrated are extensible to other zones and other GNSS arrays of varying size, number, ground distribution, and transmitter frequency.

Show moreThe redundancy of a structure refers to the extent of strength that is not considered in design. For an indeterminate structure a member failure does not necessarily induces the loss of integrity or functionality of the structure; rather it will affect its potential for safely carrying any future load. Numerous methods have been introduced in structural reliability literature to measure and implement the redundancy in design. However, in accordance with the semi-probabilistic approach of the codes which aim to develop design method providing consistent level of redundancy within the structure, the probability of failure of the structure has been proposed and is widely used as a redundancy measure. A classical method to compute the probability of failure of the structure based on failure paths is presented as a reference in this thesis. However, although extensively used, this method has major shortcomings which may lead to a misrepresentation of the structure redundancy. By using a geometric representation of members’ limit states associated with a loading regime, the research presented herein proposes an improved method for structural redundancy estimation that may be helpful to overcome problems associated with approximations and inconsistencies inherent in classics methods. Specific assumptions and/or procedures considered in the proposed method are described below. (1) An approximation is given to make the events of member failures mutually exclusive. (2) Geometric calculations are used to determine reliability indices and conditional reliability indices in order to establish closer bounds for the failure probability of individual structural members. (3) System’s failure probability is obtained using the assumption and procedures outlined in (1) and (2) above. (4) To further extend the method beyond geometrical redundancy, and to consider material redundancy, plasticity models commonly used in the structural analyses are considered in this study
