Show moreThis invention provides various biquinone and trimeric quinone derivatives. The invention also provides a method for synthesis of a multi-quinone compound including reacting a hydroxyquinone anion with a first quinone possessing a first directing group at a C-2 of the first quinone and a second directing group at a C-3 of the first quinone and obtaining a biquinone having one of the first and second directing groups at a C-3 of a first quinone monomer and a hydroxyl group at a C-3' of a second quinone monomer. The biquinone can be further reacted to obtain various biquinone derivatives or with a second hydroxyquinone anion to obtain trimeric quinone derivatives, including trimeric naphthoquinone derivatives. The biquinones and trimeric quinones of this invention demonstrate antiviral activity and can be used to treat viral infections, particularly HIV infections.

Show moreThe olfactory system is an important chemosensory system. Our identification of odorants is occurred through binding of odorant molecules with olfactory receptors in the olfactory epithelium. In zebrafish, there are four classes of olfactory receptors: trace amine-associated receptor (TAAR), odorant receptor (OR), vomeronasal type 1-like receptor (V1R-like) and vomeronasal type 2-like receptor (V2R-like). In this study, I amplified four olfactory receptors taar1a, or1016, ora5 and olfcc1, which are respectively belong to the four olfactory receptor classes, from cDNA of the zebrafish olfactory epithelium and cloned genes into a mammalian expression vector pCI. The gene sequences of these receptors were not identical as the sequences obtained from GenBank database. The possible reasons for the inconsistency of gene sequence are discussed. I further tranfected these four cloned olfactory receptors in a mammalian cell line Hana3A. Expression of all four olfactory receptors in Hana3A cells was confirmed by immunocytochemistry. In summary, these four olfactory receptors taar1a, or1016, ora5 and olfcc1 were successfully cloned into the pCI vector and can be expressed on cell membrane surface of Hana3A cells.

Show moreIt is well known that the exponential growth in popularity of wireless devices has created a demand for radio spectrum that cannot be met with current regulatory policies. Despite the difficulty in procuring access to new spectrum resources, many empirical studies have indicated that the majority of spectrum is in-fact unused in the temporal, spatial and/or spectral domains, representing an untapped wealth that must be exploited. Dynamic Spectrum Access (DSA) is a promising technology which aims to improve the efficiency of future radios and alleviate the issue of spectrum under-utilization. This dissertation utilizes the data from the IIT Spectrum Observatory to develop models of channel activity on the Land Mobile Radio (LMR) band (used for critical communication by organizations such as public safety) and shows how such models can be applied to improve the performance of DSA. We demonstrate that LMR traffic may possess multi-timescale behavior – such as clustering and dispersion over different time periods – and propose a novel statistical model to account for these observations based on a multiple emission hidden Markov model. We then used this model to design a collision constrained channel selection algorithm that can permit the re-use of licensed spectrum while minimizing interference with incumbent users. The findings in this work are primarily developed for public safety, however the techniques developed are general enough to be applied to other types of traffic possessing similar characteristics. The proposed model, in particular, is well suited for further analytic work and simulations studies in this area.

Show moreDuchenne muscular dystrophy, DMD, is a severe X-linked recessive disease that kills boys in their second or third decade of life. A related condition Becker Muscular Dystrophy, BMD, is very heterogeneous with clinical severity ranging from nearly as bad as DMD, to nearly benign. Both of these are caused by defects in the dystrophin gene, which encodes a 427 kDa cytoskeletal protein, dystrophin, which can provide a mechanical link between the cytoskeleton and the muscle membrane and stables muscle tissue. In general, DMD results from defects that eliminate all dystrophin expression, whereas BMD patients carry mutated products: modified and malfunctioned dystrophin proteins. Since DMD is invariably fatal, monogenic, has a high incidence (1:3500 male births) and afflicts a charismatic patient profile, it has been an attractive target for gene therapy. However, the huge size of the dystrophin gene (2.4 Mbp) and more importantly its processed mRNA (14.2 kb) poses a problem for viral gene therapy which is limited to payloads of about 8 kb. This has prompted efforts to reduce the size of the dystrophin protein while still maintaining functionality. A major inspiration for such efforts was the identification of a very unusual, very large deletion serendipitously found in an extremely mild BMD case. The patient identified with this defect was ambulatory at 65 year of ages. This edit removes more than half the central rod region, from exon 17 to 48, Δe17-48, and, fortunately, the size of this BMD type edit protein is available for viral vector loading. In this project, edited dystrophin with central rod region deletion (D2:22 Δe17-48) and other two related edits, D2:22 Δe17-47 (exon 48 is added in D2:22 Δe17-48) and D2:22 Δe17-49 (exon 49 is deleted from D2:22 Δe17-48), were studied. According to these results, D2:22 Δe17-47 demonstrated the best biophysical and biochemical properties, but D2:22 Δe17-48 displayed higher to protease sensitivity, and D2:22 Δe17-49 had the worst thermal stability. This work will help us to understand the factors that result in benign large scale edits, and facilitate the development of effective viral gene therapy vectors.

Show moreA method of electromagnetic heating in situ recovers liquid hydrocarbons from an oil shale formation containing kerogen in an inorganic matrix where the formation is substantially impermeable to fluids under native conditions. A block of the oil shale formation is substantially uniformly heated in situ with electromagnetic power to a temperature of about 275.degree. C. where there is pyrolysis of a portion of the kerogen to gas and shale oil at a pressure sufficient to overcome the capillary pressure of the shale oil in the matrix, thereby providing substantial fluid permeability to the formation. The gas thereupon escaping from said block and the shale oil driven thereby are recovered, thereby further increasing the permeability of the formation. The magnitude of the electromagnetic power is controlled to raise the temperature of the block relatively slowly to increase the rate of pyrolysis of the kerogen as the permeability of the formation increases to produce gas at pressures above the necessary to overcome the capillary pressure and below that at which there is substantial escape of the gas bypassing shale oil within the formation rather than driving the oil before it.

A computer is used to process input data, such as data representing frequency of the oscillatory flow within the chamber, to a flow rate passing through the spirometer. Show moreHigh temperature, short time pasteurization was used to evaluate a computer-based system for controlling the pasteurization process, acquiring data, and monitoring records. Software was used for the control of hot water temperature, flow rate through the centrifugal timing pump, and diversion of under-processed product. Three types of control strategies were conducted: single loop, cascade, and multivariable. The single loop control strategy showed the most rapid responses to temperature changes, but the temperature response curve was slowest to return to its set point. The cascade control strategy showed slower recoveries to temperature changes, but the temperature response curve was smoother. The multivariable control strategy responded slightly faster than the cascade control strategy, and the temperature response curve was slightly smoother than the cascade control strategy. The multivariable control strategy was able to control the flow diversion valve by the use of a lethality controller. The data acquisition system, used to monitor the data obtained from the high temperature, short-time pasteurization system, was within +/- 0.1 degrees C of the temperature recorded by the safety thermal limit recorder. Reliability was determined by examining the changes in the position of the flow diversion valve to identify process deviations and by comparing the changes to the event marker on circular charts. The data acquisition system was an effective alternative for monitoring the completeness of data.

The coating apparatus includes an encapsulation chamber enclosing a two-layer water-oil system for coating each islet cell with an aqueous polymeric coat. Islets together with an aqueous polymer solution are fed by a feed device that utilizes the principle of hydrodynamic focusing in order to ensure encapsulation of individual islets. The polymer in the aqueous coat is subsequently crosslinked by being exposed to laser light to produce structurally stable microcapsules of controllable thickness of the order of tens of microns. Encapsulated islets are removed from the encapsulation chamber by a valveless pump and recovered by filtration or centrifugation.

Show moreMONUMENTAL ADAPTATION: REIMAGINING THE OLD CHICAGO POST OFFICE Built in 1932, the Old Chicago Main Post Office was the largest post office in the world at three million square feet. It was designed to accommodate the extension of Congress Parkway and in 1953 the Eisenhower Expressway cut a void through the center of the building. Finally the Post Office was vacated in 1997 and remains unused to this day. This project proposes the adaptation of the Post Office into a hybrid fieldhouse that contains public recreation and retail space. This proposal keeps the Post Office public, utilizing income generated through retail to maintain the building for Chicagoans. Recreation program is inserted into voids carved out of the original building. Retail occupies the peripheral space, surrounding the voids.
