Show moreThis study examined the application of Pareto-optimal weighting schemes to real selection data from the public safety sector. Of particular interest was the evaluation of how well Pareto-optimal weighting estimates approximate observed validity and adverse impact statistics when applied to selection data. Pareto-optimal methodology was applied to entry-level selection data from two U.S. municipalities in the public safety sector. Results indicate that Pareto-optimal estimates maintain robustness when compared to real selection outcomes. Specifically, near identically-shaped Pareto fronts were obtained when inputting both raw and corrected validity estimates, suggesting suitable sensitivity to varying input parameters. Practical challenges of implementing Pareto-optimal methods as well as the implications for adopting Pareto-optimal weights are discussed.

Show moreDuring development and differentiation, the transcription regulation plays an important rule. The Polycomb Group (PcG) and the Trithorax Group (TrxG) proteins are two protein families; they act by forming multiprotein complexes that regulate transcription by modifying chromatin configurations through histone methylation. The establishment or the maintenance mistakes of the correct chromatin configuration have been connected to several diseases, including leukemia and muscular dystrophy. The activities of PcG and TrxG proteins are regulated by a special group of proteins known as Enhancers of trithorax and Polycomb (ETP). The Additional sex combs-like 2 (Asxl2) protein, which is expressed from the gene called additional sex combs like 2 (Drosophila) (Asxl2), belongs to the Enhancer of trithorax and Polycomb group (ETP). The Asxl2 protein is highly expressed in heart during both development and in adult life. In a genetic study, the Asxl2-/- mice either died as neonates with congenital heart abnormalities or displayed progressive deterioration of ventricular function. Although Asxl2 protein is important, there have been no significant studies of the Asxl2 protein’s structure. Protein functions are determined to a considerable degree by structure, so the structural studies will provide valuable information on Asxl2. Failures of protein folding into native structure generally produce inactive or toxic proteins. Misfolded proteins are believed to be the causes of some neurodegenerative diseases, including Alzheimer’s and Parkinson's diseases. ix Through investigation in silico, it is known that the Asxl2 protein contains a large native unstructured region, and this result might indicate the Asxl2 is an intrinsically disordered protein (IDP). Most IDPs are known to have regulatory roles. Owing to their lack of folded structure, the intrinsically disordered proteins display a flexible and random-coil-like conformation under physiological conditions, and have several unique features. This suggests that distinctive methods will be required to characterize their structure. In order to understand the structure of Asxl2, we have expressed and purified an Asxl2 N-terminal truncation that contains a large native unstructured region of two important domains: the HARE-HTH domain and the Asx homology domain (ASXH). We have then used Circular Dichroism (CD) as a means of investigating the structure of Asxl2. The CD spectra have been measured over a wavelength range from 250 nm to 190 nm at various temperatures, with and without Asxl2’s physiological binding partner, the ubiquity carboxyl-terminal hydrolase BAP1 (Bap1) protein in order to test the hypothesis that Asxl2 is an IDP.

Show moreStigma toward people with mental illness can have deleterious effects; employers avoid hiring them, landlords avoid renting to them, and negative attitudes are often a barrier to receiving quality health care. Literature on political ideology indicates that people tend to have fairly stable attitudes over time that are consistent with their political orientation. It is also the case that political ideology is an underlying factor of attitudes toward personal responsibility and hierarchical societal structure. Political ideology, as an underlying framework for attributing controllability through which many individuals view the world, is a demographic variable that needs to be further explored to more completely integrate variances into anti-stigma approaches. Pre-intervention data from a larger study assessing the effects of newspaper articles on attitudes about mental illness were utilized to examine the influence of political orientation on stigmatizing and affirming attitudes toward people with mental illness. Pearson product moment correlations between demographics, political orientation variables, stigmatizing attitudes, and affirming attitudes were conducted. Regression analyses were conducted to determine if political orientation or the interaction between political orientation and demographics are significantly related to stigmatizing attitudes. Results showed that political affiliation emerged as significant associated with pity, danger, blame, and anger when moderated by other demographics suggesting that endorsement of affiliation with a particular political party alone is not sufficient to emerge as related to stigmatizing attitudes, but rather varies at levels of other demographic variables.

Show moreSocial networking sites are radically transforming the way we communicate and relate to each other. They facilitate timely information exchange and give us unprecedented access to numerous sources of information on a myriad of topics. Although the information is available, there are a number of challenges that inhibit utilization of this information: Social Networks have a great volume of messages that the user must sift through to find relevant ones, messages are frequently repetitive, the information is not organized topically, and there is little context information. The information consumer (user) must take on many of the tasks traditionally performed by the information producer to get a “big picture” understanding of the topic. This thesis introduces a framework for an automated information gathering and organization system to facilitate the information consumer’s comprehension of a given topic. The framework addresses two primary components: the user interface for the system and identification of sub-topics. The framework was implemented as a research platform designed to bring these two components together and support future research in the domain.

Show moreDystrophin is a rod shaped protein consisting of amino- and carboxy-terminal binding domains linked by a large central rod composed of 24 homologous copies of the STR motif and 5 non-homologous regions termed hinges. There are no high-resolution crystallographic structures available for the STR repeats. This work yielded protein crystals and data sets of two STR fragments (d5 & d16-17). Though unable to produce a high resolution structure, this work examines the physical properties of the rod region utilizing hydrophobic moment analysis and molecular dynamics to suggest flexibility among the helices of the STR fragments. Helical shifting appears to possibly occur among the more stable STR tandems to increase the cooperation among certain adjacent fragments. A full understanding of how these properties vary along the length of the rod has implications for the engineering of these rods regions in future dystrophin therapies.
