Show moreBiocompatible scaffolds mimicking the locally aligned fibrous structure of native extracellular matrix (ECM) are in high demand in tissue engineering. In this thesis research, unidirectionally aligned fibers were generated via a home-built electrospinning system. Collagen type I, as a major ECM component, was chosen in this study due to its support of cell proliferation and promotion of neuroectodermal commitment in stem cell differentiation. Synthetic dragline silk proteins, as biopolymers with remarkable tensile strength and superior elasticity, were also used as a model material. Good alignment, controllable fiber size and morphology, as well as a desirable deposition density of fibers were achieved via the optimization of solution and electrospinning parameters. The incorporation of silk proteins into collagen was found to significantly enhance mechanical properties and stability of electrospun fibers. Glutaraldehyde (GA) vapor post-treatment was demonstrated as a simple and effective way to tune the properties of collagen/silk fibers without changing their chemical composition. With 6-12 hours GA treatment, electrospun collagen/silk fibers were not only biocompatible, but could also effectively induce the polarization and neural commitment of stem cells, which were optimized on collagen rich fibers due to the unique combination of biochemical and biophysical cues imposed to cells. Taken together, electrospun collagen rich composite fibers are mechanically strong, stable and provide excellent cell adhesion. The unidirectionally aligned fibers can accelerate neural differentiation of stem cells, representing a promising therapy for neural tissue degenerative diseases and nerve injuries.

Show moreThe study of an architecture of autonomy consists of theoretical investigations into the realm of building types where a sole use or purpose is manifest in a structure that could, site provided, be constructed. However, provisions that conventional architecture traditionally provide are not present in these explorations. Technological advancements such as indoor plumbing, electric lights, and vertical conveyance systems in the form of elevators and escalators are excluded. Platonic geometric form-making are instead thoroughly investigated, imagined, and manipulated for the purposes of creating new spatial experiences. The desired resultant is an architecture of singularity, an architecture of fantastical projection. Through a series of two theoretical ritual-based investigations, three-dimensional form manipulation and construction of proportioned scale models, the essence of elements that compose a spatial experience contributed to a collection of metaphorical tools by which the designer may use to build a third imagined reality: the re-imagination of the archetypal museum. A building whose purpose is not solely to house ancient objects in a near hermetically-sealed environment, free of temperature, humidity and ultra-violet light aberrations, but is a re-imagined. A structure meant to engage the presence of two seemingly divergent communities: the local patron/visitor and the extreme distant denizen. This paper also examines key contemporary global artists’ work and their contributions to the fragmentation / demolition of architectural assemblages for thepurposes of re-evaluating the familiar vernacular urban landscape while criticallypositioning the rôle of both the artifact and gallery in shaping contemporary audience’s museum experiences. The power of the internet and live-camera broadcasting of images utilizing both digital image recording and full-scale screen-projections enable the exploration of “transporter-type” virtual-reality experiences: the ability to inhabit an art work’s presumed original in situ location, while remaining in Chicago as a visitor within a vernacular multi-tenant masonry structure: vacated, evicted, and deconstructed for the purposes of displaying art amidst a new urbane ruin. The complexities of this layered experience is meant to simultaneously displace and interrupt a typical set of so-called a priori gallery expectations while providing the expectant simulacrum that video cameras and screens provide, whetting a contemporary patron’s appetite.positioning the rôle of both the artifact and gallery in shaping contemporary audience’s museum experiences. The power of the internet and live-camera broadcasting of images utilizing both digital image recording and full-scale screen-projections enable the exploration of “transporter-type” virtual-reality experiences: the ability to inhabit an art work’s presumed original in situ location, while remaining in Chicago as a visitor within a vernacular multi-tenant masonry structure: vacated, evicted, and deconstructed for the purposes of displaying art amidst a new urbane ruin. Show moreModel-based controllers for a bench scale autothermal tubular packed-bed reactor have been formulated using the Internal Model Control (IMC) approach. The Structural Dominance Analysis technique has been used in developing the reduced-order models. Controller performance at robust and sensitive steady states have been assessed through simulations and experiments. Both PI and model-based controllers can regulate reactor operation at robust steady states, but only third order IMC controllers are able to regulate reactor operation at the sensitive steady state.

Show moreWith the improvements in sensor technologies and image processing algorithms, computer vision has become a major tool for robots to recognize and gauge their surroundings. For instance, the Kinect sensor can be used as an excellent depth camera for indoor navigation. However, there exist situations that need recognition and spatial interpretation of the environment using limited hardware resources. The Kinect is not suitable for outdoor use, while LIDAR is too large and expensive to be installed on an autonomous miniature surveillance drone. Therefore, the use of a single camera is the only feasible option for many embedded applications. To perform SfM (structure from motion) by using single camera is challenging due to the complexity of 3D mapping. Feature detection and mapping is the very fist step to perform SfM. To be more specific, matched feature points are used as anchors cross images or frames. Without such matched feature points, most SfM method will not be able to generate reliable results; moreover, instead of using frames from videos as inputs, most feature detectors and matching strategies are designed for SfM applications using images as inputs. Therefore, this thesis will discuss how to detect feature points from video and match them effectively. Image projection and SfM fundamentals will be introduced in this thesis as well.

Show moreAssessing progress in different construction activities at the end of every payment period is time consuming and requires specialized personnel employed by the contractor and the owner. Automatic progress control that requires a minimum amount of human involvement could reduce the time spent on this activity, reduce the number of personnel used, reduce the cost involved, reduce disagreements between contractor and owner, and add to the overall efficiency of project management. Attempts have been made in the past to resolve this issue using image processing and other techniques but the results have not been satisfactory. A new attempt was made to set up a system that can assess progress control with minimum human input and the results are presented in this paper. The experiment made use of laser scanning technology and was conducted both in laboratory conditions and construction sites. The initial results from laboratory condition appear to be promising but there are still obstacles to surmount. The system is robust and accurate in laboratory conditions and constitutes proof of concept. Improvements are made to accelerate the registration process of multiple scans, to reduce the noise data, to recognize objects of irregular shape, and to assess the practicality and economic feasibility of such a system when applying this system in real construction sites. Show moreAsxl2 is a chromatin factor that regulates histone H3 methylation and H2A deubiquitination. Previous research in our lab has shown that Asxl2 is an important regulator of mammalian heart development and function. To facilitate the characterization of Asxl2, we proposed to generate an affinity reagent for the detection of Asxl2 proteins in vivo and in vitro. A recombinant GST-Asxl2 fusion protein in E. coli was produced that contained GST and the N-terminal 390 amino acids of Asxl2, separated by a Precision protease cleavage site. Asxl2 N1-390 was purified from GSTAsxl2 N1-390 and the purified polypeptide was biotinylated and used to select a phagedisplay antibody library. Finally, six antibodies were produced that exhibit various degrees of affinity with the recombinant Asxl2 N1-390 in ELISA assay. In the future the antibodies will be tested for their ability to detect endogenous Asxl2 proteins in western blot assay. This would facilitate the study of the role of Asxl2 in heart development and function.

Show moreA two-dimensional nonlinear distributed-parameter optimal state estimation algorithm has been developed and applied to an exothermic packed bed catalytic reactor. Temperature and concentration profiles were estimated from limited temperature and/or concentration measurements. Orthogonal collocation was used to solve the differential sensitivity and filtering equations. Both simulations and on-line experiments with a pilot scale packed bed reactor for the partial oxidation of methanol have been used to study the effects of measurement choice, model parameter errors, and unmeasured process disturbances. Excellent tracking of the reactor profiles was achieved with only one or two properly placed temperature sensors.
